SARS-CoV-2 (COVID-19) Nucleic Acid Detection Kit (Fluorescent RT-PCR)


Weiyun SARS-CoV-2 (COVID-19) Nucleic Acid Detection Kit (Fluorescent RT-PCR) is a TaqMan probe real-time fluorescent PCR test product to qualitatively detect the nucleic acids of ORF1ab, N and E genes to SARS-CoV-2 in throat swab and sputum of patients with respiratory infection symptoms and their close contacts. The test results are for clinical reference only and cannot be used as the solo basis for diagnosis. Operators should have professional training experience in gene amplification or molecular biology testing, and have relevant qualifications for laboratory operations. The laboratory should have reasonable biosafety precautions and protective measures.


Test Principle

The kit selects the conserved sequences of ORF1ab, N and E genes of SARS-CoV-2 for the designing of specific primers and probes, and performs real-time fluorescent PCR test on viral RNA after nucleic acid purification.

 

During PCR amplification, the probe binds to the template, and the 5'-end reporter group of the probe is cleaved by the Taq enzyme (5'→3' exonuclease activity), thereby moving away from the quenching group to generate fluorescent signals.

 

The real-time amplification curve is automatically plotted based on the detected fluorescence signal, and the sample Ct value is calculated. FAM, VIC and Texas Red fluorophores are labeled to ORF1ab gene, N gene and E gene probes in reaction solution. In this way, qualitative detection of the above three genes of SARS-CoV-2 can be performed simultaneously in one test.

 

The kit provides an internal control (human gene fragment labeled with a Cy5 fluorescent group) to monitor the possible inhibitory factors in the extraction process and reaction system to avoid false negative results.
